Glutathione is a small tripeptide made of glutamic acid, cysteine, and glycine. Its cysteine thiol group allows reversible oxidation and reduction, making it central to cellular redox chemistry. The reduced form, often abbreviated GSH, predominates inside most cells, while the oxidized disulfide form, GSSG, forms when two GSH molecules react. The ratio of GSH to GSSG is widely used as an indicator of oxidative stress in laboratory research, though it does not by itself diagnose a clinical condition.
Biosynthesis occurs in two ATP-dependent steps. The enzyme glutamate-cysteine ligase joins glutamate and cysteine, forming gamma-glutamylcysteine; glutathione synthetase then adds glycine to produce the complete tripeptide. Because the peptide bond from glutamate uses the gamma-carboxyl group, glutathione resists digestion by many ordinary peptidases. Tissues vary in synthesis capacity, and the liver generally contains high concentrations relative to many other organs. This uneven distribution contributes to organ-specific differences in redox buffering and affects how experimental results are interpreted across tissue types.
Glutathione participates in detoxification reactions, amino acid transport, and the maintenance of protein thiols. It serves as a cofactor for several enzymes, including glutathione peroxidases and glutathione S-transferases. In research literature, altered glutathione status appears in studies of aging, infection, metabolic stress, and environmental exposure. Whether low glutathione is a cause, consequence, or marker of such conditions often remains unresolved. Direct measurement in blood or tissue provides a snapshot, but results depend on sample handling, timing, and the method used.
Common analytical approaches include enzymatic recycling assays, high-performance liquid chromatography, and mass spectrometry. The enzymatic recycling assay uses glutathione reductase and a colorimetric or fluorometric reagent to amplify signal, which gives good sensitivity for total glutathione. Chromatographic methods can separate GSH from GSSG and related thiols, while mass spectrometry offers structural confirmation and multiplexing. Each approach has different requirements for calibration, internal standards, and validation. No single method captures every form of glutathione in every matrix.
Storage recommendations for glutathione reagents usually specify a cool, dry, dark environment because the thiol oxidizes in air and light. Solid material is often kept desiccated at low temperature, while solutions are prepared fresh or stored frozen in aliquots. Repeated freeze-thaw cycles can accelerate degradation, and metal ions can catalyze oxidation. Quality control may include purity assays, water content, and identity confirmation. Stability limits are method-specific, so a stated shelf life applies only to defined conditions and packaging.

Synthesis occurs in two ATP-dependent steps. The enzyme glutamate-cysteine ligase joins glutamate and cysteine to form gamma-glutamylcysteine, and glutathione synthetase adds glycine. The first step is rate-limiting and is influenced by cysteine availability and feedback inhibition by GSH. Breakdown involves gamma-glutamyl transferase and subsequent peptidases, which release constituent amino acids for reuse. Because turnover differs among tissues, measurements from blood, plasma, and tissues are not directly interchangeable. Research continues to clarify how compartment-specific pools are regulated in health and disease.
Glutathione is a tripeptide composed of glutamate, cysteine, and glycine. It contains an unusual gamma-glutamyl bond between glutamate and cysteine, which resists cleavage by many peptidases. The reduced form, GSH, carries a thiol group on cysteine and is the dominant intracellular form in most cells. Its structure allows it to participate in redox reactions and to serve as a sulfur donor. The oxidized form, GSSG, consists of two GSH molecules joined by a disulfide bond.
In cells, glutathione helps maintain the reducing environment of the cytosol and supports enzymes that counteract reactive oxygen species. It acts as a cofactor for glutathione peroxidases, which reduce hydrogen peroxide and lipid peroxides, and for glutathione S-transferases, which conjugate electrophiles. The ratio of GSH to GSSG is often used as an indicator of oxidative stress, although the ratio can vary by compartment and cell type. Glutathione also stores cysteine, an amino acid that can be limiting for protein synthesis and antioxidant defense.
For solid glutathione reagents, storage at low temperature and protection from moisture and light are typical precautions. Aqueous solutions can oxidize over time, and pH affects stability; alkaline conditions generally promote thiol oxidation. Some protocols prepare fresh solutions, while others use antioxidants or chelators to limit metal-catalyzed oxidation. Purity and counterion content can vary among commercial preparations, affecting concentration calculations. Certificates of analysis and validated assays help verify identity and purity.
Measuring glutathione in biological samples requires attention to oxidation, because GSH can convert to GSSG after sample collection. Blood and plasma samples are often treated with acid or alkylating agents to preserve the reduced form. Without stabilization, apparent GSH concentrations can fall while GSSG rises. Differences in sample type, handling delay, and deproteinization method can produce results that are not comparable across studies. Reporting preanalytical details is therefore important for interpreting findings.
Glutathione supports detoxification by conjugating reactive electrophiles through glutathione S-transferases. The resulting conjugates are processed and exported, often after further metabolism. It also stores cysteine, transports amino acids across membranes through the gamma-glutamyl cycle, and assists in the maturation of iron-sulfur clusters and some prostaglandins. In plants, animals, and many microbes, the molecule appears in similar roles, but concentrations vary enormously between tissues. Liver, kidney, and red blood cells tend to contain high amounts, while blood plasma contains much less.
Glutathione is a small tripeptide built from glutamate, cysteine, and glycine. The peptide bond between glutamate and cysteine uses the gamma-carboxyl group, a linkage that resists ordinary peptidases. Cells make it in two ATP-dependent steps: gamma-glutamylcysteine synthetase joins glutamate and cysteine, then glutathione synthetase adds glycine. The pathway is feedback-inhibited by glutathione itself, so intracellular levels tend to stay within a narrow range. Because cysteine is often limiting, sulfur amino acid supply influences how much glutathione a cell can produce.
In its reduced form, glutathione carries a sulfhydryl group that can donate electrons. This property lets it act as a major cellular antioxidant and redox buffer. Glutathione peroxidase uses it to reduce hydrogen peroxide and lipid peroxides, while glutathione reductase regenerates the reduced form using NADPH. The ratio of reduced glutathione to glutathione disulfide is widely used as an indicator of oxidative stress, though the ratio changes with compartment, cell type, and sample handling. Oxidized glutathione can also form mixed disulfides with proteins, affecting their activity.
However, benzodiazepines have been used clinically to manage the adverse psychological effects of psychedelics, for instance in clinical studies and in the emergency department. A clinical trial of psilocybin and midazolam coadministration found that midazolam clouded the effects of psilocybin and impaired memory of the experience. Benzodiazepines might interfere with the therapeutic effects of psychedelics, such as sustained antidepressant effects. Some serotonergic psychedelics, for instance dimethyltryptamine (DMT) and 5-MeO-DMT, are highly susceptible substrates for monoamine oxidase (MAO), specifically MAO-A, and hence can be greatly potentiated by monoamine oxidase inhibitors (MAOIs). An example of this is ayahuasca, in which plants containing both DMT and harmala alkaloids acting as MAOIs such as harmine and harmaline are combined. This allows DMT to become orally active and to have a much longer duration of action than usual. The 2C psychedelics, such as 2C-B, 2C-I, and 2C-E, are also substrates of both MAO-A and MAO-B, and may likewise be greatly potentiated by MAOIs. Examples of MAOIs that may potentiate psychedelics behaving as MAO-A and/or MAO-B substrates include phenelzine, tranylcypromine, isocarboxazid, moclobemide, and selegiline. Combination of MAO-substrate psychedelics with MAOIs can result in overdose and serious toxicity, including death. Other psychedelics, such as LSD, are not substrates of MAO and are not potentiated by MAOIs.
=== 2016 AFL season === The CAS verdict meant that Essendon's squad was suddenly twelve players short for the 2016 season. The club received permission to recruit ten top-up players from lower levels on contracts lasting until 31 October 2016 to supplement its list. The top-ups had to have either been on an AFL list in 2014 or 2015, with no more than one player taken from any single state-level club; or have been a VFL-listed player from Essendon's own reserves team. An allowance for the top-ups was made in Essendon's salary cap. The top-up players were: Ryan Crowley, James Kelly, Matthew Stokes, Matt Dea, James Polkinghorne, Jonathan Simpkin, Mark Jamar, Sam Grimley, Nathan Grima and Sam Michael. The four other AFL clubs with suspended Essendon players on their lists (Melbourne, Port Adelaide, St Kilda and the Western Bulldogs) were allowed to upgrade a rookie to the senior list, which would not normally be allowed to cover a suspended player, but were not granted top-up players. The twelve suspended players still at Essendon continued to be paid about 95% of their salaries by the club during their suspensions. Stewart Crameri continued to be paid by the Western Bulldogs, with the Bulldogs suing Essendon to recover the cost. Melbourne, Port Adelaide and St Kilda ceased payments to their suspended players, those players suing Essendon privately for their salaries.

=== Detection methods === Mass spectrometry (MS) is used to identify and quantify metabolites after optional separation by GC, HPLC, or CE. GC-MS was the first hyphenated technique to be developed. Identification leverages the distinct patterns in which analytes fragment. These patterns can be thought of as a mass spectral fingerprint. Libraries exist that allow identification of a metabolite according to this fragmentation pattern . MS is both sensitive and can be very specific. There are also a number of techniques which use MS as a stand-alone technology: the sample is infused directly into the mass spectrometer with no prior separation, and the MS provides sufficient selectivity to both separate and to detect metabolites. For analysis by mass spectrometry, the analytes must be imparted with a charge and transferred to the gas phase. Electron ionization (EI) is the most common ionization technique applied to GC separations as it is amenable to low pressures. EI also produces fragmentation of the analyte, both providing structural information while increasing the complexity of the data and possibly obscuring the molecular ion. Atmospheric-pressure chemical ionization (APCI) is an atmospheric pressure technique that can be applied to all the above separation techniques. APCI is a gas phase ionization method, which provides slightly more aggressive ionization than ESI which is suitable for less polar compounds.

==== Cyanopyrrolidines ==== Cyanopyrrolidines have two key interactions to the DPP-4 complex: 1. Nitrile in the position of the scissile bond of the peptidic substrate that is important for high potency. The nitrile group forms reversible covalent bonds with the catalytically active serine hydroxyl (Ser630), i.e. cyanopyrrolidines are competitive inhibitors with slow dissociation kinetics. 2. Hydrogen bonding network between the protonated amino group and a negatively charged region of the protein surface, Glu205, Glu206 and Tyr662. All cyanopyrrolidines have basic, primary or secondary amine, which makes this network possible but these compounds usually drop in potency if these amines are changed. Nonetheless, two patent applications unveil that the amino group can be changed, i.e. replaced by a hydrazine, but it is claimed that these compounds do not only act via DPP-4 inhibition but also prevent diabetic vascular complications by acting as a radical scavenger.
=== Social issues === In 2023, Celia Medrano described Bukele's positions on social issues as "flexible" ("flexibles") and a "liquid ideology" ("ideología líquida"). She explained that Bukele changes his positions to appease as many voters as possible and to gauge public opinion on issues such as same-sex marriage and abortion. Bukele stated in 2014 that he was an ally of the LGBT community, supported their civil rights, and opposed discrimination against LGBT individuals. In August 2021, Bukele proposed constitutional reform to legalize same-sex marriage in El Salvador. The proposal would have changed text in the constitution that defined marriage as being between "a man and a woman" ("hombre y mujer") to defining marriage as between "spouses" ("cónyuges"), and would have prohibited discrimination based on sexual orientation. The earliest Bukele's proposal could have gone into effect would have been 2027, since it would have to be approved by two consecutive sessions of the Legislative Assembly. Bukele stated the following month that the proposed constitutional reform would not legalize same-sex marriage, posting on Facebook that the original text would remain intact. In March 2024, Bukele stated that his government would remove "all traces" of "gender ideologies in schools and colleges". In June 2024, Bukele fired 300 bureaucrats from the ministry of culture for promoting policies that were "incompatible" with his emphasis on "patriotic and family values". El Salvador has one of the world's strictest abortion laws, banning it in all circumstances with no exceptions.
OHPH, also known as hydroxyprogesterone enanthate (OHPE), as well as 17α-hydroxyprogesterone heptanoate or 17α-hydroxypregn-4-ene-3,20-dione 17α-heptanoate, is a synthetic pregnane steroid and a derivative of progesterone and 17α-hydroxyprogesterone. It is a progestogen ester; specifically, it is the C17α heptanoate (enanthate) ester of 17α-hydroxyprogesterone. Analogues of OHPH include the more well-known medications hydroxyprogesterone acetate and hydroxyprogesterone caproate (hydroxyprogesterone hexanoate). The C3 benzilic acid hydrazone of OHPH, hydroxyprogesterone heptanoate benzilic acid hydrazone (OHPHBH), is known and has been studied in animals. In terms of chemical structure, OHPH is very similar to hydroxyprogesterone caproate, differing from it only in having one additional carbon in its fatty acid ester chain.

Authorities in Italy have investigated a 'Ndrangheta mafia clan accused of trafficking and illegally dumping nuclear waste. According to a whistleblower, a manager of the Italy state energy research agency Enea paid the clan to get rid of 600 drums of toxic and radioactive waste from Italy, Switzerland, France, Germany, and the United States, with Somalia as the destination, where the waste was buried after buying off local politicians. Former employees of Enea are suspected of paying the criminals to take waste off their hands in the 1980s and 1990s. Shipments to Somalia continued into the 1990s, while the 'Ndrangheta clan also blew up shiploads of waste, including radioactive hospital waste, sending them to the sea bed off the Calabrian coast. According to the environmental group Legambiente, former members of the 'Ndrangheta have said that they were paid to sink ships with radioactive material for the last 20 years. In 2008, Afghan authorities accused Pakistan of illegally dumping nuclear waste in the southern parts of Afghanistan when the Taliban were in power between 1996 and 2001. The Pakistani government denied the allegation.
Naturally occurring ytterbium (70Yb) is composed of seven stable isotopes: 168Yb, 170Yb–174Yb, and 176Yb, with 174Yb being the most abundant (31.90% natural abundance). 30 radioisotopes have been characterized, with the most stable being 169Yb with a half-life of 32.014 days, 175Yb with a half-life of 4.185 days, and 166Yb with a half-life of 56.7 hours. All of the remaining radioactive isotopes have half-lives that are less than 2 hours, with the majority of them being less than 20 minutes. This element also has 18 meta states, with the most stable being 169mYb (half-life 46 seconds). The known isotopes of ytterbium range from 149Yb to 189Yb. The primary decay mode before the most abundant stable isotope, 174Yb is electron capture giving thulium isotopes; the primary mode after is beta emission giving lutetium isotopes. Of interest to modern quantum optics, the different ytterbium isotopes follow either Bose–Einstein statistics or Fermi–Dirac statistics, leading to different behavior in optical lattices.
Irwin was the first to observe that if the size of the plastic zone around a crack is small compared to the size of the crack, the energy required to grow the crack will not be critically dependent on the state of stress (the plastic zone) at the crack tip. In other words, a purely elastic solution may be used to calculate the amount of energy available for fracture. The energy release rate for crack growth or strain energy release rate may then be calculated as the change in elastic strain energy per unit area of crack growth, i.e.,
On 15 April 2023, the country's paramilitary Rapid Support Forces launched attacks against al-Burhan's government, claiming to capture key government sites. Al-Burhan claimed these sites were still under the control of his forces, and that fighting was ongoing. The clashes continued until at least 21 April with occasional ceasefires. Al-Burhan was pinned down at SAF headquarters in Khartoum until August 2023, when a military operation enabled him to evacuate to Port Sudan, where he has been based since then. The Sudanese military allegedly received support from Ukraine. Ukraine has also allegedly sent troops to fight in Omdurman. Their rival, the Rapid Support Forces, had reportedly received support from Libya and the United Arab Emirates as well as Russia's Wagner Group. The foreign involvement in the regional war could have a devastating effect on Sudan. Al-Burhan survived an assassination attempt while attending a military graduation ceremony in Gebeit, in July 2024. Five people were killed in the drone attack. In August 2025, a military official in Sudan confirmed that al-Burhan's envoy, Al-Sadiq Ismail, visited Israel in a push to speed the process of formal normalization between both countries through the Abraham Accords, which has been delayed due to the war. It was reported that Sudan wanted Israeli support following the capture of Khartoum by the SAF in exchange for Sudan's recognition.

GSH is the reduced form of glutathione, with a free thiol group on cysteine. GSSG is the oxidized disulfide form, created when two GSH molecules become linked. The two forms exist together, and their balance is often reported as the GSH/GSSG ratio in laboratory studies.
Glutathione is synthesized inside cells from amino acids rather than being classified as an essential dietary nutrient. Dietary sources can provide glutathione or its precursors, but digestion and absorption alter what reaches tissues. Research continues on how dietary intake relates to cellular glutathione levels.
The liver has high glutathione concentrations and uses the compound in conjugation and antioxidant reactions. These reactions are relevant to the processing of drugs, pollutants, and normal metabolic byproducts. Studies often examine liver glutathione as a marker of oxidative stress or detoxification capacity.
Pre-analytical handling, extraction chemistry, and detection method all influence reported glutathione values. Oxidation during sample processing can shift the measured GSH/GSSG ratio. Standardized protocols and reference materials help reduce, but do not eliminate, these differences.
