Interpreting glutathione measurements requires attention to pre-analytical variables. The GSSG concentration in a sample can rise artificially during storage or processing, making the GSH/GSSG ratio unreliable if not controlled. Reference ranges vary by specimen type, assay, and population, so comparisons across studies are difficult. Plasma glutathione is low and sensitive to hemolysis, while whole blood reflects primarily erythrocyte content. Many studies measure total glutathione rather than the reduced and oxidized forms separately, which limits conclusions about redox status.
Accurate measurement of glutathione begins with careful sample handling. Because GSH oxidizes rapidly to GSSG, samples must be processed quickly or frozen immediately. Acid precipitation with metaphosphoric acid or perchloric acid is common; it lowers pH, precipitates proteins, and helps preserve the reduced form. Chelating agents such as EDTA can limit metal-catalyzed oxidation. For whole blood, hemolysis releases glutathione from erythrocytes, so plasma and serum values differ substantially from whole blood values.
Several analytical methods can quantify glutathione, including high-performance liquid chromatography (HPLC) with UV or fluorescence detection for separating GSH and GSSG. Liquid chromatography-tandem mass spectrometry (LC-MS/MS) offers higher specificity and sensitivity, often detecting nanomolar concentrations. The enzymatic recycling assay, often called the Tietze method, measures total glutathione by coupling reduction of GSSG to a colorimetric or fluorometric readout. Capillary electrophoresis and electrochemical detection are also used in specialized laboratories. Each method has distinct advantages and limitations regarding throughput, cost, and susceptibility to interference.
Enzymatic recycling assays provide a complementary approach for total glutathione. In these methods, glutathione reductase reduces oxidized glutathione while a thiol-reactive reagent, such as 5,5'-dithiobis(2-nitrobenzoic acid), produces a colored product. The reaction cycles between reduced and oxidized forms, amplifying the signal. Spectrophotometric or fluorometric detection can then estimate concentration. Distinguishing reduced glutathione from glutathione disulfide often requires separate aliquots, masking agents, or chromatographic separation, and the choice affects reported values.
Storage conditions strongly influence glutathione stability. The solid reduced form is commonly kept desiccated at or below minus twenty degrees Celsius, protected from light and moisture. Aqueous solutions are less stable because the thiol group reacts with dissolved oxygen, and oxidation accelerates at neutral or alkaline pH. Acidic solutions and oxygen-free handling can slow degradation, but repeated freeze-thaw cycles should be avoided. Researchers often verify concentration before use, because apparent losses can arise from oxidation or water uptake.

Glutathione participates in detoxification reactions, amino acid transport, and the maintenance of protein thiols. It serves as a cofactor for several enzymes, including glutathione peroxidases and glutathione S-transferases. In research literature, altered glutathione status appears in studies of aging, infection, metabolic stress, and environmental exposure. Whether low glutathione is a cause, consequence, or marker of such conditions often remains unresolved. Direct measurement in blood or tissue provides a snapshot, but results depend on sample handling, timing, and the method used.
Glutathione is a small tripeptide made of glutamic acid, cysteine, and glycine. Its cysteine thiol group allows reversible oxidation and reduction, making it central to cellular redox chemistry. The reduced form, often abbreviated GSH, predominates inside most cells, while the oxidized disulfide form, GSSG, forms when two GSH molecules react. The ratio of GSH to GSSG is widely used as an indicator of oxidative stress in laboratory research, though it does not by itself diagnose a clinical condition.
For solid glutathione, storage conditions affect shelf life. The reduced form is typically kept cool, dry, and protected from air and light. Moisture can promote oxidation, while elevated temperatures accelerate degradation. Suppliers often specify storage at or below freezing, sometimes under inert gas. Solutions are less stable than powders and may require preparation shortly before use. Buffers and chelating agents can slow oxidation, but they do not eliminate it. Published stability data vary with matrix, pH, and container.
Quality control for glutathione focuses on identity, purity, and oxidation state. Certificates of analysis may report assay value, water content, and the presence of GSSG or other impurities. Chromatographic purity is often expressed as a percentage of peak area. Reference standards help laboratories compare results across instruments and batches. Because glutathione is a small, polar molecule, separation from cysteine, gamma-glutamylcysteine, and related thiols can be challenging. Verification often combines more than one analytical technique.
=== Biomedical applications === By examining the behaviours of the molecular 'switch' peptides, more information about interactions between proteins and, more significantly, the pathogenesis of some protein conformational diseases can be obtained. These diseases include scrapie, kuru, Huntington's, Parkinson's and Alzheimer's. Self-assembling and surfactant peptides can be used as targeting delivery systems for genes, drugs and RNAi. Research has already shown that cationic dipeptides NH2-Phe-Phe-NH2 nanovesicles, which are about 100 nm in diameter, can be absorbed into cells through endocytosis and deliver oligonucleotides into the cell; this is one example of how peptide nanostructure can in used in gene and drug delivery. It is also envisaged that water-soluble molecules and biological molecules would be able to be delivered to cells in this way. Self-assembling LEGO peptides can form biologically compatible scaffolds for tissue repair and engineering, which should be of great potential, as a large number of diseases cannot be cured by small molecule drugs; a cell-based therapy approach is needed and peptides could potentially play a huge role in this. Cyclic peptide nanotubes formed from self-assembly can act as ion channels, which form pores through the cell membrane and cause cellular osmotic collapse. Peptide can be designed to preferentially form on bacterial cell membranes and thus these tubes can perform as antibacterial and cytotoxin agents.
Sertraline, sold under the brand name Zoloft among others, is an antidepressant medication of the selective serotonin reuptake inhibitor (SSRI) class used to treat major depressive disorder, generalized anxiety disorder, social anxiety disorder, obsessive–compulsive disorder (OCD), panic disorder, and premenstrual dysphoric disorder. Although also having approval for post-traumatic stress disorder (PTSD), findings indicate it leads to only modest improvements in symptoms associated with this condition. The drug shares the common side effects and contraindications of other SSRIs, with high rates of nausea, diarrhea, headache, insomnia, mild sedation, dry mouth, and sexual dysfunction, but it appears not to lead to much weight gain, and its effects on cognitive performance are mild. In some patients, sexual dysfunction may persist even after the drug is discontinued, a condition known as post-SSRI sexual dysfunction; regulatory agencies including the European Medicines Agency and Health Canada have recommended that sertraline's product labeling warn of this risk. Similar to other antidepressants, the use of sertraline for depression may be associated with a mildly elevated rate of suicidal thoughts in people under the age of 25 years old. It should not be used together with monoamine oxidase inhibitors (MAOIs): this combination may cause serotonin syndrome, which can be life-threatening in some cases. Sertraline taken during pregnancy is associated with an increase in congenital heart defects in newborns.
The brief existence of an mRNA molecule begins with transcription, and ultimately ends in degradation. During its life, an mRNA molecule may also be processed, edited, and transported prior to translation. Eukaryotic mRNA molecules often require extensive processing and transport, while prokaryotic mRNA molecules do not. A molecule of eukaryotic mRNA and the proteins surrounding it are together called a messenger RNP.
=== Biophysical limitations === The tree of the African locust bean requires "between 0-300 metres of altitude, a mean annual rainfall of between 400-700 millimetres and a mean annual temperature of about 24-28°C." It prefers well-drained, thick clay soils but can also be found on shallow, thin sandy soils.

==== Inhalation and smoking ==== Results from epidemiological studies indicate that the risk of lung cancer increases with exposure to residential radon. One well known potential source of error in these studies is smoking, which is the main risk factor for lung cancer. In the US, cigarette smoking is estimated to cause 80% to 90% of all lung cancers. Radon, like other known or suspected external risk factors for lung cancer, is a threat for smokers and former smokers. According to the EPA, the risk of lung cancer for smokers is significantly higher when they are exposed to radon due to the synergistic effects of radon with smoking. For this population, about 62 people in a total of 1,000 will die of lung cancer, compared to 7 people in a total of 1,000 for people who have never smoked. A 2005 review article by Darby and colleagues analyzing 13 different case studies across Europe stated: "It is not appropriate to talk simply of a risk from radon in homes. The risk is from smoking, compounded by a synergistic effect of radon for smokers. Without smoking, the effect seems to be so small as to be insignificant." According to Darby, there is a difference in risk for the histological subtypes of lung cancer and radon exposure. Small-cell lung carcinoma, which has a high correlation with smoking, has a higher risk after radon exposure. For other histological subtypes, such as adenocarcinoma, the type that primarily affects non-smokers, the risk from radon appears to be lower.
He had wanted to say: "You sound as though from the tomb", but instead he told Thomas that he sounded like Louis Armstrong. Later, Thomas went drinking with Reitell at the White Horse and, feeling sick again, returned to the hotel. Feltenstein came to see him three times that day, administering the cortisone secretant ACTH by injection (a tropic peptide hormone produced and secreted by the anterior pituitary gland) and, on his third visit, half a grain (32.4 milligrams) of morphine sulphate. This amount was three times more than was medically appropriate and was, by affecting Thomas's breathing and consequently the oxygen supply to his brain, the precipitating cause of the coma from which he would never awake. After Reitell became increasingly concerned and telephoned Feltenstein for advice he suggested she get male assistance, so she called upon the painter Jack Heliker, who arrived before 11 pm. At midnight on 5 November, Thomas's breathing became more difficult and his face turned blue. Reitell phoned Feltenstein who arrived at the hotel at about 1 am, and called for an ambulance. It then took another hour for the ambulance to arrive at St. Vincent's, even though it was only a few blocks from the Chelsea. Thomas was admitted to the emergency ward at St Vincent's Hospital at 1:58 am. He was comatose, and his medical notes state that "the impression upon admission was acute alcoholic encephalopathy damage to the brain by alcohol, for which the patient was treated without response".
== Discovery == Originally, there was no common name for the skeleton of coicenal A-C. In 2013, Quan-xin Wang and his research team in Beijing discovered the structure of coicenal D by reacting the carbon skeletons of coicenal A and coicenal B with acetyl chloride at room temperature. This novel finding and resultant publication prompted the official classification of these 4 diterpenes as coicenal A-D. Wang's team began their study to search for new secondary metabolites from a solid culture of Bipolaris coicis. They used high-performance liquid chromatography with diode array detection and searched for characteristic UV maximal absorption at 280 nm. This investigation was inspired by previous discoveries of secondary metabolites in the Bipolaris family by other researchers.
== Pharmacokinetics == With the once-monthly formulation of paliperidone palmitate, the time to peak is 13 days and the elimination half-life is 25 to 49 days; for the 3-month formulation, the time to peak is 30 to 33 days and the half-life is 84 to 95 days (deltoid muscle) or 118 to 139 days (gluteal). These data come from a 2021 review whose authors included employees of Janssen Research & Development, the manufacturer of paliperidone palmitate. The peak-to-trough ratio of paliperidone palmitate at steady state ranges from 1.56 to 1.70 with the 1- and 3-month formulations. No pharmacokinetic data for the 6-month formulation has been released as of January 2021. However, a 2025 case report described a patient who still had measurable plasma paliperidone concentrations in the low therapeutic range 886 days (2.5 years) after her last injection, indicating that in some individuals the drug may be released from the depot far longer than the predicted half‑life. The pharmacokinetic profile was originally derived from a 2009 pooled analysis of 18,530 paliperidone concentration samples from 1,795 subjects across six Phase I trials and five Phase II and III trials funded by Johnson & Johnson Pharmaceutical Research and Development, LLC. Invega Sustenna, Invega Trinza, and Invega Hafyera all utilize nanoparticle technology. The NanoCrystal® technology used in all three long-acting paliperidone products is manufactured by Alkermes plc.

=== 1950–1980: Pivot to pharmaceutical research and global expansion === Due to price declines for penicillin, Pfizer searched for new antibiotics with greater profit potential. Pfizer discovered oxytetracycline in 1950, and this changed the company from a manufacturer of fine chemicals to a research-based pharmaceutical company. Pfizer developed a drug discovery program focused on in vitro synthesis to augment its research in fermentation technology. In 1959, the company established an animal health division with a 700-acre (280 ha) farm and research facility in Terre Haute, Indiana. By the 1950s, Pfizer had established offices in Belgium, Brazil, Canada, Cuba, Mexico, Panama, Puerto Rico, and the United Kingdom. In 1960, the company moved its medical research laboratory operations out of New York City to a new facility in Groton, Connecticut. In 1980, Pfizer launched Feldene (piroxicam), a prescription anti-inflammatory medication that became Pfizer's first product to reach $1 billion in revenue. In 1965, John Powers, Jr. became chief executive officer (CEO) of the company, succeeding John McKeen. As the area surrounding its Brooklyn, NY plant fell into decline in the 1970s and 1980s, the company formed a public-private partnership with New York City that encompassed the construction of low- and middle-income housing, the refurbishment of apartment buildings for the homeless and the establishment of a charter school. In 1972, Edmund T. Pratt Jr. became CEO of the company, succeeding John Powers, Jr.
The two major inactive metabolites are the N-dealkylation products (the carboxylic acid ID-20219 and the piperazine ID-11614), and a norbornane hydroxylated derivative of ID-20219 (ID-20220). Of lurasidone and its metabolites circulating in the blood, the native drug makes up 11%, the main active metabolite 4%, and the inactive carboxylic acids 24% and 11%, respectively. Several dozen metabolites have been identified altogether.:59–61 Biological half-life is given as 18 hours or 20 to 40 hours in different sources. 80% or 67% of a radiolabelled dose was recovered from the feces, and 9% or 19% from the urine.
In 2009, in an article for The Atlantic entitled "The Revenge of Karl Marx", Hitchens frames the late-2000s recession in terms of Marx's economic analysis and notes how much Marx admired the capitalist system that he called for the end of, but says that Marx ultimately failed to grasp how revolutionary capitalist innovation was. Hitchens was an admirer of Che Guevara, yet in an essay written in 1997, he distanced himself from Che, and referred to the mythos surrounding him as a "cult". In 2004 he re-emphasized his positive view of Che, commenting that "[Che's] death meant a lot to me and countless like me at the time. He was a role model, albeit an impossible one for us bourgeois romantics insofar as he went and did what revolutionaries were meant to do – fought and died for his beliefs." He continued to regard Leon Trotsky and Vladimir Lenin as great men, and the October Revolution as a necessary event in the modernisation of Russia. In 2005, Hitchens praised Lenin's creation of "secular Russia" and his attacks on the Russian Orthodox Church, describing the church's power as "absolute warren of backwardness and evil and superstition". In God is not Great, Hitchens called Marxism his "own secular faith" that "has been shaken and discarded, not without pain." He referred to his period of Marxist faith as "when I was a Marxist." According to Andrew Sullivan, his last words were "Capitalism, downfall."
Dermal macrophages are macrophages in the skin that facilitate skin homeostasis by mediating wound repair, hair growth, and salt balance. Their functional role in these processes is the mediator of inflammation. They can acquire an M1 or M2 phenotype to promote or suppress an inflammatory response, thereby influencing other cells' activity via the production of pro-inflammatory or anti-inflammatory cytokines. Dermal macrophages' ability to acquire pro-inflammatory properties also potentiates them in cancer defence. M1 macrophages can suppress tumour growth in the skin by their pro-inflammatory properties. However, M2 macrophages support tumour growth and invasion by the production of Th2 cytokines such as TGFβ and IL-10. Thus, the exact contribution of each phenotype to cancer defence and the skin's homeostasis is still unclear. Dermal macrophages are native immune cells mainly located in the dermis of the skin. They consist of two distinct populations: yolk-sac derived precursors or circulating monocytes. They are tissue-resident phagocytes that facilitate microbial debris clearance in the skin. Dermal macrophages belong to the mononuclear phagocyte system that serves a vital role in the innate immunity of the skin. They are also a type of antigen-presenting cells (APCs) that can mediate the infiltration of immune cells during an immune response, suggesting dermal macrophages' influence on both the innate and adaptive immune systems. However, their antigen-presenting ability is relatively lower than dendritic cells (DCs) and Langerhans cells (LCs) in the skin.

Glutathione oxidizes quickly when cells are disrupted or when samples sit at room temperature. Rapid processing or immediate freezing minimizes the conversion of GSH to GSSG. This step helps ensure that the measured ratio reflects the original biological state.
The Tietze assay is an enzymatic recycling method that measures total glutathione. It uses glutathione reductase to reduce GSSG back to GSH, which then reacts with a chromogen or fluorophore. The reaction cycles repeatedly, amplifying the signal for detection.
Yes, but the choice of blood fraction matters. Plasma or serum contains low glutathione levels and is easily affected by hemolysis. Whole blood mainly reflects the high glutathione content of erythrocytes, so results from different fractions are not directly comparable.
Chromatographic methods can separate the two forms before detection. Enzymatic assays often measure total glutathione first and then use a separate procedure to estimate the oxidized fraction. The difference between total and oxidized amounts provides an indirect estimate of the reduced form.
