Common analytical approaches include enzymatic recycling assays, high-performance liquid chromatography, and mass spectrometry. The enzymatic recycling assay uses glutathione reductase and a colorimetric or fluorometric reagent to amplify signal, which gives good sensitivity for total glutathione. Chromatographic methods can separate GSH from GSSG and related thiols, while mass spectrometry offers structural confirmation and multiplexing. Each approach has different requirements for calibration, internal standards, and validation. No single method captures every form of glutathione in every matrix.
Storage recommendations for glutathione reagents usually specify a cool, dry, dark environment because the thiol oxidizes in air and light. Solid material is often kept desiccated at low temperature, while solutions are prepared fresh or stored frozen in aliquots. Repeated freeze-thaw cycles can accelerate degradation, and metal ions can catalyze oxidation. Quality control may include purity assays, water content, and identity confirmation. Stability limits are method-specific, so a stated shelf life applies only to defined conditions and packaging.
Measuring glutathione requires attention to oxidation during sample handling, because GSH in biological samples can convert to GSSG or form mixed disulfides with proteins after collection. Acidic extraction, rapid cooling, and chelating agents are commonly used to limit such changes. Analytical methods usually distinguish free reduced glutathione, total glutathione, and protein-bound forms. Because these forms have different stability and reactivity, reported values depend heavily on the preparation protocol. No single preparation is universally suitable for every biological matrix or analytical goal.
Several techniques are used for quantification. Enzymatic recycling assays rely on glutathione reductase and a colorimetric or fluorescent readout, offering sensitivity for total glutathione. High-performance liquid chromatography can separate GSH from GSSG and other thiols, often with UV, fluorescence, or electrochemical detection. Mass spectrometry provides structural confirmation and can quantify low-abundance species when paired with separation. Each approach has trade-offs in specificity, throughput, and equipment requirements, so method selection depends on the research question and available instrumentation.
| Property | Value | Notes |
|---|---|---|
| Typical storage temperature | -20 °C or below | For solid reagent and frozen aliquots; protect from moisture and light. |
| Common analytical method | HPLC with UV or fluorescence detection | Separates GSH and GSSG after derivatization or direct detection. |
| Alternative method | LC-MS/MS | Provides high specificity and can quantify multiple thiols. |
| Total glutathione assay | Enzymatic recycling | Uses glutathione reductase and a chromogen or fluorogen. |
| Key stability risk | Oxidation to GSSG | Air, light, and trace metals promote conversion. |
Enzymatic recycling assays provide a complementary approach for total glutathione. In these methods, glutathione reductase reduces oxidized glutathione while a thiol-reactive reagent, such as 5,5'-dithiobis(2-nitrobenzoic acid), produces a colored product. The reaction cycles between reduced and oxidized forms, amplifying the signal. Spectrophotometric or fluorometric detection can then estimate concentration. Distinguishing reduced glutathione from glutathione disulfide often requires separate aliquots, masking agents, or chromatographic separation, and the choice affects reported values.
Storage conditions strongly influence glutathione stability. The solid reduced form is commonly kept desiccated at or below minus twenty degrees Celsius, protected from light and moisture. Aqueous solutions are less stable because the thiol group reacts with dissolved oxygen, and oxidation accelerates at neutral or alkaline pH. Acidic solutions and oxygen-free handling can slow degradation, but repeated freeze-thaw cycles should be avoided. Researchers often verify concentration before use, because apparent losses can arise from oxidation or water uptake.
Measuring glutathione in biological samples requires attention to oxidation and matrix effects. High-performance liquid chromatography with ultraviolet or fluorescence detection can separate reduced and oxidized forms after derivatization. Liquid chromatography with tandem mass spectrometry offers higher specificity and can quantify glutathione alongside related thiols. Because glutathione can oxidize during sample handling, many protocols use rapid acidification with metaphosphoric acid or sulfosalicylic acid. Internal standards help correct for losses during extraction and analysis.
Measuring glutathione in biological samples requires attention to oxidation, because GSH can convert to GSSG after sample collection. Blood and plasma samples are often treated with acid or alkylating agents to preserve the reduced form. Without stabilization, apparent GSH concentrations can fall while GSSG rises. Differences in sample type, handling delay, and deproteinization method can produce results that are not comparable across studies. Reporting preanalytical details is therefore important for interpreting findings.
Common analytical approaches include enzymatic recycling assays, high-performance liquid chromatography, and mass spectrometry. Enzymatic recycling measures total glutathione after converting GSSG back to GSH, while separation methods can quantify GSH and GSSG separately. Derivatization may be used to improve detection or stability during analysis. LC-MS/MS offers high specificity and can distinguish glutathione from related thiols and adducts. Each method has different sensitivity, throughput, and susceptibility to interference, so method selection depends on the study question and sample matrix.
For solid glutathione reagents, storage at low temperature and protection from moisture and light are typical precautions. Aqueous solutions can oxidize over time, and pH affects stability; alkaline conditions generally promote thiol oxidation. Some protocols prepare fresh solutions, while others use antioxidants or chelators to limit metal-catalyzed oxidation. Purity and counterion content can vary among commercial preparations, affecting concentration calculations. Certificates of analysis and validated assays help verify identity and purity.
Glutathione is synthesized in two ATP-dependent steps. First, gamma-glutamylcysteine synthetase links glutamate and cysteine; second, glutathione synthetase adds glycine to form the complete tripeptide. The pathway is feedback-inhibited by GSH itself, which helps maintain steady intracellular levels. Tissues vary widely in glutathione content, with the liver typically containing the highest concentrations, followed by the kidneys, lungs, and erythrocytes. Because cysteine is often limiting, its availability influences synthesis rates, and regulation of this pathway varies by cell type.
Glutathione serves as a cofactor for several enzymes, including glutathione peroxidase and glutathione S-transferase. These enzymes help reduce hydrogen peroxide and lipid peroxides, and they conjugate reactive electrophiles for excretion. The molecule also acts as a reservoir for cysteine, an amino acid that is prone to oxidation. In addition, glutathione participates in the metabolism of nitric oxide, leukotrienes, and prostaglandins. Its roles extend to cell signaling, apoptosis, and the regulation of protein function through S-glutathionylation.
Glutathione is a tripeptide composed of glutamate, cysteine, and glycine, and it is the most abundant non-protein thiol in most living cells. The reduced form, GSH, carries a sulfhydryl group that can donate electrons, while the oxidized form, GSSG, forms when two GSH molecules link via a disulfide bond. The balance between these two forms helps define the cellular redox environment, and their ratio is often used as an indicator of oxidative stress. Because the sulfhydryl group is reactive, glutathione participates in many cellular processes, including detoxification and protein regulation.
Some of the earliest human remains found in the Americas, Luzia Woman, were found in the area of Pedro Leopoldo, Minas Gerais, and provide evidence of human habitation going back at least 11,000 years. The earliest pottery ever found in the Western Hemisphere was excavated in the Amazon basin of Brazil and radiocarbon dated to over 8,000 years ago (6000 BC). The pottery was found near Santarém and provides evidence that the region supported a complex prehistoric culture. The Marajoara culture flourished on Marajó in the Amazon delta from AD 400 to 1400, developing sophisticated pottery, social stratification, large populations, mound building, and complex social formations such as chiefdoms. Around the time of the Portuguese arrival, the territory of present day Brazil had an estimated Indigenous population of 7 million people, mostly semi-nomadic, who subsisted on hunting, fishing, gathering, and migrant agriculture. The population comprised several large Indigenous ethnic groups (e.g., the Tupis, Guaranis, and Gês. The Tupi people were subdivided into the Tupiniquins and Tupinambás. Before the arrival of the Europeans, the boundaries between these groups and their subgroups were marked by wars that arose from differences in culture, language and moral beliefs. These wars also involved large-scale military actions on land and water, with cannibalistic rituals on prisoners of war. While heredity had some weight, leadership was a status more won over time than assigned in succession ceremonies and conventions.

Riboflavin, also known as vitamin B2, is a vitamin found in food and sold as a dietary supplement. It is essential to the formation of two major coenzymes, flavin mononucleotide and flavin adenine dinucleotide. These coenzymes are involved in energy metabolism, cellular respiration, and antibody production, as well as normal growth and development. The coenzymes are also required for the metabolism of niacin, vitamin B6, and folate. Riboflavin is prescribed to treat corneal thinning, and taken orally, may reduce the incidence of migraine headaches in adults. Riboflavin deficiency is rare and is usually accompanied by deficiencies of other vitamins and nutrients. It may be prevented or treated by oral supplements or by injections. As a water-soluble vitamin, any riboflavin consumed in excess of nutritional requirements is not stored; it is either not absorbed or is absorbed and quickly excreted in urine, causing the urine to have a bright yellow tint. Natural sources of riboflavin include meat, fish and fowl, eggs, dairy products, green vegetables, mushrooms, and almonds. Some countries require its addition to grains. In its purified, solid form, it is a water-soluble yellow-orange crystalline powder. In addition to its function as a vitamin, it is used as a food coloring agent. Biosynthesis takes place in bacteria, fungi and plants, but not animals. Industrial synthesis of riboflavin was initially achieved using a chemical process, but current commercial manufacturing relies on fermentation methods using strains of fungi and genetically modified bacteria.

== Business career == Newsom and a group of investors created the company PlumpJack Associates L.P. on May 14, 1991. The group started the PlumpJack Winery in 1992 with the financial help of his family friend Gordon Getty. PlumpJack was the name of an opera written by Getty, who invested in 10 of Newsom's 11 businesses. Getty told the San Francisco Chronicle that he treated Newsom like a son and invested in his first business venture because of that relationship. According to Getty, later business investments were because of "the success of the first". One of Newsom's early interactions with government occurred when Newsom resisted the San Francisco Department of Public Health's requirement to install a sink at his PlumpJack wine store. The Health Department argued that wine was a food and required the store to install a $2,700 sink in the carpeted wine shop on the grounds that the shop needed the sink for a mop. When Newsom was later appointed supervisor, he told the San Francisco Examiner: "That's the kind of bureaucratic malaise I'm going to be working through." The business grew to an enterprise with more than 700 employees. The PlumpJack Cafe Partners L.P. opened the PlumpJack Café, also on Fillmore Street, in 1993. Between 1993 and 2000, Newsom and his investors opened several other businesses that included the PlumpJack Squaw Valley Inn with a PlumpJack Café (1994), a winery in Napa Valley (1995), the Balboa Café Bar and Grill (1995), the PlumpJack Development Fund L.P.
=== PIP1 and PIP2 === The 3-D structures of the open and closed forms of plant aquaporins, PIP1 and PIP2, have been solved (PDB: 4JC6). In the closed conformation, loop D caps the channel from the cytoplasm and thereby occludes the pore. In the open conformation, loop D is displaced up to 16 Å, and this movement opens a hydrophobic gate blocking the channel entrance from the cytoplasm. These results reveal a molecular gating mechanism which appears conserved throughout all plant plasma membrane aquaporins. In plants it regulates water intake/export in response to water availability and cytoplasmic pH during anoxia.
Clitoris: Made up of the root, glans and body and is usually retracted into a prepuce. Inside the clitoris of many non-human placentals is the baubellum, a small bone that possibly has origins in copulation. In horses and dogs, the clitoris is contained in clitoral fossa, which is a small pouch of tissue. Labia: A small, thin pair of lip-like structures that protect the vestibule. They are known as the labia vulvae in carnivorans and ungulates and as the labia minora in primates. The labia majora only exist in primates (including humans). Afrotherians do not have distinguishable labia. Vestibule/vulvar opening: In humans, other great apes, and some rodents, the vestibule is a flat and short external space that contains separate urethral and vaginal openings. In most other placentals, the urethra and vagina join as an internal vestibule (urogenital sinus), hence both urine and offspring exit through an orifice called the vulvar opening. During estrus, the clitoris of a mare (female horse) everts as the labia contracts by opening and closing. This is colloquially known as "winking". Throughout the menstrual cycle, some female primates' vulvar and anal regions will swell (sexual swelling) to attract a male, though the fundamental reason for this function is up for debate. The vulva of a spotted hyena has a large clitoris known as a pseudo-penis for copulating, giving birth and urinating, as well as fused labia (pseudo-scrotum). This can make it difficult to correctly sex the species.

The small vessels in the skin affected are located in the superficial dermis and include arterioles (small arteries carrying blood to capillaries), capillaries, and venules (small veins receiving blood from capillaries). In general, immune complexes deposit in vessel walls leading to activation of the complement system. C3a and C5a, proteins produced from the complement system, attract neutrophils to the vessels. Once activated, neutrophils then release preformed substances, including enzymes, causing damage to vessel tissue. Evidence of this process can be seen with a sample of removed skin tissue, or biopsy, viewed under a microscope. Neutrophils are seen surrounding blood vessels and their debris within vessel walls, causing fibrinoid necrosis. This finding on histological examination is termed "leukocytoclastic vasculitis". Considering the wide range of potential causes leading to cutaneous small vessel vasculitis, there are subtle variations in the underlying pathophysiology for each cause. For example, medications are metabolized to smaller molecules that can attach to proteins in the blood or vessel walls. The immune system senses these altered proteins as foreign and produces antibodies in efforts to eliminate them from the body. A similar process occurs with infectious agents, such as bacteria, in which antibodies target microbial components.

Pre-analytical handling, extraction chemistry, and detection method all influence reported glutathione values. Oxidation during sample processing can shift the measured GSH/GSSG ratio. Standardized protocols and reference materials help reduce, but do not eliminate, these differences.
Total glutathione typically refers to the combined amount of reduced glutathione and glutathione disulfide, expressed in glutathione equivalents. Assays that measure total glutathione do not distinguish GSH from GSSG unless a separation step is included. Researchers often pair a total assay with a specific GSSG measurement to estimate the redox ratio.
Glutathione reference standards are generally stored cold, dry, and protected from light. Weighed portions should be prepared promptly and used within validated stability windows. Purity and water content can affect the accuracy of calibration curves.
Common methods include enzymatic recycling assays, liquid chromatography, and mass spectrometry. Many protocols separate reduced glutathione from its oxidized disulfide form before detection.
